Abstract:
 Miniaturizing analysis systems reduces both cost and time for analyzing processes in different biological and environmental aspects. One of the most significant forms of this miniaturization is point of care system or lab on a chip. The objective of this thesis is to introduce a self-powered miniaturized analysis system. Capillary forces are used to manipulate the liquids through the system. DNA amplification using polymerase chain reaction (PCR) was demonstrated using this system as a proof of its concept.
Firstly, an introduction to the new research field of miniaturizing analysis systems is introduced including its applications and development. The fabrication of microfluidic devices is also briefed. Detailed discussion of capillary action theory and capillary based microfluidics is then presented to pave the way for the design of the microfluidic system.
A detailed description of DNA capillary analysis system components is given. Each component is discussed separately. A new design of the capillary trigger valve is discussed. After that, the design calculations of the micromixer are presented. The design of the micro capillary pump is then described stating the different types of the pumps. Lastly, numerical simulation was used to develop a new correlation of the permeability of cylindrical micropillar arrays. The new correlation was used in the design calculations of the capillary pump.
Finally, device fabrication and experimental characterization of the system are discussed. The device was fabricated in silicon using deep reactive ion etching. A brief introduction to DNA amplification is given. Fluidic characterization was performed by comparing the measured values of the flow velocity with the calculated values. Direct PCR from blood experiment was demonstrated to prove the concept of the analysis system.
